This presentation provides a practical overview of how Tosoh resins can be applied in oligonucleotide purification. It begins with the current landscape of oligonucleotide therapeutics, common structural modifications, and the main purification routes used in process development. The document compares reversed-phase chromatography, anion-exchange chromatography, and hydrophobic interaction chromatography for their suitability in different oligonucleotide purification tasks. It further presents application examples for TSKgel SuperQ-5PW, Toyopearl SuperQ / GigaCap Q, and TSKgel Phenyl-3PW in the purification of DNA oligonucleotides, phosphorothioate-modified oligonucleotides, morpholino oligonucleotides, and siRNA. In addition, it highlights the potential of HIC for online DMT-on / DMT-off separation and on-column DMT cleavage, and suggests that oligonucleotide downstream processing is gradually shifting from RPC-based routes toward HIC + AEC combinations for improved efficiency and reduced solvent usage.
